Investigation of COI Gene Variations in Domestic Populations of Spodoptera litura in Korea

Abstract

The tobacco cutworm, Spodoptera litura, is a significant agricultural pest in Korea. Recent evidence indicates its population has expanded in response to climate change. In this study, a total of 12,540 adult S. litura individuals were collected in 2024 from 135 cities and counties across nine regions including Gyeongbuk (GB), Gyeongnam (GN), Daegu (DG), Gyeonggi (GG), Gangwon (GW), Chungbuk (CB), Chungnam (CN), Jeonbuk (JB), Jeonnam (JN), and Jeju. Among all collections, GN accounted for the highest proportion (44%) of individuals, while Jeju showed the lowest (0.9%). To assess regional genetic variation, mitochondrial cytochrome c oxidase subunit I (COI) gene sequences were analyzed among the collected populations. The COI gene analysis revealed similar single nucleotide polymorphism patterns across regions, with most individuals exhibiting a C→T substitution at position 406. However, additional regional variations were detected including a C→T substitution at position 407 in Uljin (GB), Pocheon (GG), Taebaek (GW), and Wanju (JB); a C→T substitution at position 59 in Uljin (GB); and an A→G substitution at position 34 in Hakjung (DG). Five haplotypes (Hap1-Hap5) were identified from the nationwide dataset. These findings provide the first nationwide assessment of COI haplotype variation in Korean populations of S. litura and suggest limited genetic differentiation among geographical populations.
