Brain Transcriptomic Analysis of Fall Armyworm, Spodoptera frugiperda under Cold Exposure
Abstract
To elucidate the molecular mechanisms underlying cold tolerance in Spodoptera frugiperda, a transcriptomic analysis was conducted on brain tissue following exposure to 4°C as cold temperature. A total of 266,051 unigenes were identified, among which 6,917 genes were significantly differently expressed under cold temperature. More genes were down-regulated than up-regulated overall in response to cold. Notably, several genes associated with cold stress were identified including those involved in protein stability, membrane protection, and energy metabolism. Among the differentially expressed genes (DEGs), Hsp and PEDS1 genes exhibited over 50-fold up-regulation, while RN7SK gene showed a dramatic 175-fold down-regulation in the cold-exposed group. Quantitative RTPCR validation of major DEGs confirmed expression trends consistent with RNA-Seq analysis. These findings provide a deeper molecular understanding of cold adaptation physiology in insects and offer potential molecular markers for studying cold resistance and environmental adaptation mechanisms in pest management.
